氧自由基诱发离体大鼠胃粘膜细胞的损伤 被引量:1

OXYGEN RADICAL-INDUCED INJURY OF ISOLATED GASTRIC MUCOSAL CELLS OF THE RAT

摘要 本实验采用Pronase-EDTA法分离大鼠胃粘膜细胞,以黄嘌氧化酶(XO)-黄嘌呤(X)系统产生氧自由基,观察了氧自由基对离体孵育的胃粘膜细胞的损伤作用。结果表明,XO—X系统可使细胞存活率下降和乳酸脱氢酶(LDH)漏出增加,二者呈显著的负相关关系,其变化幅度与XO—X的作用时间或XO浓度呈依从关系;O_2清除剂超氧化物歧化酶和OH清除剂甘露醇及二甲亚砜均可减轻XO—X诱发的细胞存活率下降及LDH漏出增加。表明,O_2和OH在XO—X诱发的离体胃粘膜细胞损伤中起主要作用。 The present work observed the role of oxygen radicals on the injury of gastric mucosal cells which were isolated from the rat with pronase-EDTA method. Oxygen radicals were produced by xanthine oxidase (XO)-xanthine (X) system. The results showed, that XO-X system could reduce the viability of the cells and increase the lactate dehydrogenase (LDH) leakage from the cells, that the variations of both had a negative correlationship and were dependent on the duration of XO-X acting on the cells and on the concentration of XO, and that superoxide dismutase (a scavenger of superoxide radi- cal) and mannitol or dimethyl sulfoxide (two scavengers of hydroxy radical) might inhibit the reduc- tion of cell viability and the LDH leakage from the cells induced with XO-X system. The above results suggested that superoxide and hydroxy radicals played a main role in the injury of isolated gastric mucosal cells induced with XO-X system.
