摘要
目的研究miR-139—3p在胃癌组织和癌旁组织中的表达水平及其对AGS和SGC-7901胃癌细胞株的增殖和凋亡的作用。方法采用qRT—PCR检测38例患者胃癌组织与癌旁组织中miR-139—3p的表达情况,并分析miR-139—3p的表达与胃癌临床病理特征之间的关系;利用miR-139—3p抑制剂下调胃癌细胞株中miR-139—3p的表达,MTT法检测对AGS和SGC-7901细胞增殖的影响,流式细胞仪检测细胞凋亡情况。结果miR-139—3p在胃癌组织中表达高于癌旁组织(P〈0.01),其表达增高与肿瘤大小和部位相关(均P〈0.05);下调miR-139—3p的表达能够抑制胃癌细胞的增殖、促进细胞凋亡(均P〈005)。结论miR-139—3p在胃癌组织中表达增高,可能参与胃癌的发生、发展过程。
Objective To investigate the effect of miR-139-3p on proliferation and apoptosis in gastric cancer. Methods The expression of miR-139-3p in gastric cancer and pericancerous tissues was examined by Real-time PCR. The correlation between miR-139-3p expression and clinicopathologic features of gastric cancer was analyzed. The cultured gastric cancer AGS and SGC-7901 cells were treated with miR-139-3p Inhibitor; the cell proliferation was detected by M]q- assay and apopto- sis was analyzed by flow cytometry. Results The expression of miR-139-3p increased in gastric cancer compared with peri- cancerous tissues (P〈0.05). Expression of miR-139-3p was associated with tumor size (P〈0.05) and location (P〈0.05). Down-regulation of miR-139-3p inhibited proliferation and promoted apoptosis of gastric cancer AGS and SGC-7901 cells. Conclusion miR-139-3p is up-regulated in gastric cancer, which might be involved in the development of gastric cancer.
