摘要 目的:测定不同产地刺五加不同部位的多糖含量。方法:采用微波提取法提取刺五加中的多糖,考察微波提取时间、料液比、提取功率3个因素对多糖提取率的影响,确定最佳提取条件,并采用苯酚-硫酸法测定其含量。结果:微波提取法提取刺五加中多糖的最佳条件为提取功率700W,料液比1∶30,作用时间120s;刺五加根部及茎部多糖含量均以本溪产最高,穆枝产次之,珲春产最低。结论:用微波提取法提取刺五加中的多糖具有快速、高效、节能、不破坏有效成分等特点,具有良好的应用前景;苯酚-硫酸法简便、快速、准确,可作为刺五加多糖的含量测定方法;不同产地、不同部位刺五加药材的多糖成分的含量不同。 Objective: The polysaccharide contents of different parts of radix acanthopanacis senticosi from differ- ent sources were studied. Methods: Polysaccharides of radix acanthopanacis senticosi were extracted by microwave. The optimal conditions of polysaccharide extraction were determined including the microwave extraction time, the ra- tio of material to liquid and extraction power, which were the influencing factors to the extraction rate of polysaccha- rides. The polysaccharide content was determined by the phenol-sulfuric acid method. Results: The best conditions to extract polysaccharides from radix acanthopanacis senticosi were at 1:30 ratio of material to liquid under 700 W mi- crowave power for 120 s. Radix acanthopanacis senticosi from Benxi showed the highest polysaccharide contents in both root and stem, and less and least polysaccharide contents showed from Muzhi and Hunchun respectively. Con- clusion: The microwave extraction of polysaccharides from radix acanthopanacis senticosi exhibited the advantages of better extraction rate, high efficiency, less energy consumption, and without destroying the effective contents. This method showed a good application prospect for the content extraction. The phenol-sulfuric acid method was proved simple, rapid and accurate for the content determination of polysaccharides. The polysaccharide contents of different parts of radix acanthopanacis senticosi fi'om different sources were varied.
