摘要
目的:确定黄粉虫黄酮提取的最佳工艺。方法:以黄粉虫为原料,采用超声波水提取黄粉虫黄酮。通过紫外分光光度法对黄粉虫黄酮进行鉴定,提取液在400~700nm波长扫描测得最大吸光度在510nm波长处,确定含有黄酮类化合物。同时,在单因素试验基础上,应用Box-Behnken试验设计和响应面分析法,探讨液料比、超声时间、浸提温度、超声功率对黄酮提取量的影响。结果:超声波水提取黄粉虫黄酮的最佳工艺条件为液料比18.34:1(mL/g)、超声时间30.06min、浸提温度58.43℃、超声功率380.62W,实际测量值为11.47mg/g,理论预测值为11.58mg/g。结论:实际优化的工艺条件与理论预测拟合程度高。
Objective: To establish optimum conditions for extracting flavonoids from Tenebrio molitor. Methods: Flavonoids were extracted from Tenebrio molitor by ultrasonic-assisted aqueous extraction. The obtained extract was found to have maximum absorption at 510 nm and thus identified to contain flavonoids. Response surface methodology based on Box-Behnken experimental design was used to explore the effect of liquid-to-solid ratio, ultrasonic treatment time, temperature and ultrasonic power on extraction efficiency. Results: The optimum extraction conditions were 18.34:1 (mL/g), 30.06 min, 58.43 ~C and 380.62 W for liquid-to-solid ratio, ultrasonic treatment time, temperature and ultrasonic power, respectively. Under these conditions, the actual and predicted yields of flavonoids from Tenebrio molitor were 11.47 mg/g and 11.58g mg/g, respectively. Conclusion: The optimized conditions for flavonoid extraction from Tenebrio molitor are reliable.
