Response Surface Methodology for the Optimization of Enzymatic Extraction of Resveratrol Grape Pomace with Cellulase

摘要 为优化纤维素酶酶解提取白藜芦醇工艺,在单因素试验的基础上进行响应面试验,选择酶添加量、酶解时间和酶解温度为自变量,以白藜芦醇含量为响应值。结果表明:3个因素对白藜芦醇提取效果影响的主次顺序依次为酶添加量>酶解时间>酶解温度,酶添加量为极显著影响因素,酶解时间和酶解温度影响不显著。确定纤维素酶酶解处理山葡萄渣白藜芦醇提取工艺的最佳条件为酶添加量1.30mg/g、酶解时间1.48h、酶解温度55.49℃,在此条件下提取得到的白藜芦醇含量为0.746mg/g,是优化前提取得到的白藜芦醇含量的1.15倍。 The extraction of resveratrol from grape pomace by cellulase hydrolysis was optimized using response surface methodology. The extraction yield of resveratrol was extremely significantly influenced by enzyme dosage but not significantly impacted by time or temperature. The optimum conditions for enzymatic extraction of resveratrol from grape pomace were found to be hydrolysis at 55.49℃ for 1.48 h with an enzyme/substrate ratio of 1.30 mg/g. The extraction yield of resveratrol under these conditions was 0.746 mg/g, which was 1.15-fold higher than before optimization.
