摘要
目的:研究瓜蒌薤白颗粒与组方药材瓜蒌、薤白有效部位特征图谱色谱峰的相关性。方法:以瓜蒌薤白颗粒醋酸乙酯部位和正丁醇部位为研究对象,通过HPLC/PDAD联用建立瓜蒌薤白颗粒及其组方药材有效部位特征图谱,以渐进窗口正交投影分析法进行色谱峰匹配,利用均值和中位数建立共有模式,采用相关系数和夹角余弦计算瓜蒌薤白颗粒及其组方药味与共有模式间的相似程度。结果:瓜蒌薤白颗粒及瓜蒌的醋酸乙酯部位间有22个共有峰,利用相关系数计算平均相似度:取均值为0.766 1,取中位数为0.732 3,利用夹角余弦计算平均相似度:取均值为0.847 2,取中位数为0.829 8;瓜蒌薤白颗粒及薤白的正丁醇部位间有10个共有峰,利用相关系数计算平均相似度:取均值为0.789 3,取中位数为0.775 6,利用夹角余弦计算平均相似度:取均值为0.829 0,取中位数为0.816 0。结论:瓜蒌薤白颗粒有效部位主要色谱峰与其组方药味间有较好的相关性,采用该法可帮助确定药效部位色谱峰的归属,进一步分析瓜蒌薤白颗粒有效部位化学组成及来源,为瓜蒌薤白颗粒的质量控制提供理论依据。
OBJECTIVE To study the correlation between fingerprint peaks of GualomXiebai granules and the effective fraction of the relevant herbs in its prescription. METHODS By taking the ethyl acetate extraction and the n-butanol extraction in Gualou-Xiebai granules as the research object, the characteristic spectrum for Gualou-Xiebai granules and the effective fraction of the relevant herbs in its prescription was established by HPLC-PDAD combination, peaks was matched with the asymptotic window orthogonal projection analytical method, the common pattern was established by the mean and the median, and the similarity degree between GualowXiebai granules and the effective fraction of the relevant herbs in its prescription was calculat- ed by the correlation coefficient method and the included angle cosine method. RESULTS There were 22 common peaks between GualowXiebai granules and the ethyl acetate extraetion of Gualou, the average similarity degree was calculated by the correlation coefficient method., as the value of mean was 0. 766 1, and the value of the median was 0. 732 3, and the average similarity degree was calculated by the included angle cosine method: as the value of mean was 0. 847 2, and the value of the median was 0. 829 8; there were 10 common peaks between Gualou-Xiebai granules and the n-butanol extraction of Xiebai, the average similarity degree was calculated by the correlation coefficient method., as the value of mean was 0. 789 3, and the value of the median was 0. 775 6, and the average similarity degree was calculated by the included angle cosine method: as the value of mean was 0. 829 0, and the value of the median was 0. 816 0. CONCLUSION Main peaks had good correlation between the effective fraction of Gualou-Xiebai granules and the ingredients in its prescription, by using this method, the assignment of chromatographic fingerprints for the effective fraction was selected and the chemical components or sources of effective fraction of Gualou-Xiebai granules were further analyzed, this method can provide theoretical basis for quality control of Gualou-Xiebai granules.
