摘要
建立了HPLC同时测定茶叶中10种有效成分含量的分析方法。采用20%乙醇为提取溶剂,以Elite-SinoChrom ODS-BP(5μm,250 mm×4.6 mm i.d.)为色谱柱,柱温30℃。以甲醇(A)-1%乙酸(B)为流动相梯度洗脱,采用二极管阵列检测器进行检测,检测波长280 nm。在优化的色谱条件下,10种有效成分在0.16~1740 mg/L范围内线性关系良好,相关系数(r)不低于0.9998,检出限为0.82~48.50 ng(S/N=3)。方法可为评价茶叶及其相关产品的质量控制提供参考。
A precise and sensitive method was developed for the simultaneous determination of ten polyphenollC compounds in tea leaves by high performance liquid chromatography (HPLC). The ten compounds were extracted by 20% ethanol solution from the tea powder, and separated on an Elite-SinoChrom ODS-BP column (5 p.m,250 nm × 4. 6 mmi. d. ) at 30 ℃. A gradient program was carried out at a flow rate of 1.0 mL/min using methanol and 1% acetic acid solution as the eluent. The diode assay detection was used for qualitative and quantitative analysis and the detection wavelength was set at 280 nm. The results indicated that the chromatographic peaks of 10 compounds and their corresponding spectra were ideal under the optimal conditions. The calibration curves of ten analytes were linear in the range of 0. 16 - 1740 mg/L, with correlation coefficients more than 0. 998. The limits of detection (LODs) of ten analytes were in the range of 0. 82 48.50 ng ( S/N = 3 ). The method could provide reference for evaluating the quality of tea and other related products.
