牛膝醇提物体内诱导兔骨关节炎模型软骨修复的病理学观察 被引量:37

The Pathological observation of Alcohol extractive of Achyranthis bidentatae in vivo inducing the model of Osteoarthritis of Rabbit to repair cartilage

摘要 目的:观察牛膝醇提物对兔骨关节炎模型软骨病理学改变的影响。方法:将36例实验兔分为空白组(4只)和骨关节炎模型组(32只),对骨关节炎模型组动物采用关节伸直拉制动法进行造模。将造模成功的26只实验兔随机分为骨关节炎模型对照组、骨关节炎模型治疗组,各13只,分别予蒸馏水、牛膝醇提物灌胃4周后进行关节粘连改善度测定以及软骨病理标本大体观察、Pelletier评分、HE染色、Mankin’s评分、Ⅱ型胶原表达免疫组化检测。结果:灌胃4周后,骨关节炎模型对照组关节粘连改善度为(69.17±5.13)°,骨关节炎模型治疗组为(87.08±5.32)°,两组比较差异有统计学意义(P<0.05);前者Pelletier评分为(2.83±0.30),后者为(2.75±0.33),两组比较差异无统计学意义(P>0.05);前者软骨细胞数量明显减少,基质染色变浅,Mankin’s评分(6.91±0.53),后者软骨细胞数量增多,软骨细胞呈卵圆形,潮线结构恢复正常,Mankin’s评分为(3.67±0.47),两组比较差异有显著统计学意义(P<0.01);Ⅱ型胶原表达阳性百分率比较,前者为(32.05±13.78)%,后者为(44.45±12.62)%,两组比较差异有显著统计学意义(P<0.01)。结论:牛膝醇提物能改善关节粘连,有效刺激软骨细胞增殖,增加软骨Ⅱ型胶原表达,恢复软骨基质成分,稳定潮线结构,修复软骨损伤。 Objective :To observe pathological changes of Alcohol extractive of Achyranthis bidentatae in vivo inducing the model of Oste- oarthritis of Rabbit to repair cartilage. Methods :36 cases of rabbits were divided into 4 cases of blank group,32 cases of model of steoarthritis group ,and For steoarthritis group rabbits use joint stretched pull braking method made die. Will build die success only 26 experimental rab- bits were randoMy divided into the Control group of model of steoarthritis, the treatment group of model of steoarthritis, 13 cases each,which were Gavaged respectively by distilled water, AEAB, and after 4 weeks, to detect the degree of improvement of joint adhesion and General ob- servation,Pelletier score,HE staining,Manking score,type II collagen expression detected by immunohistochemistry of specimens of Cartilage~ Pathology. Results: After 4 weeks Gavaging, the Control group of model of steoarthritis :the degree of improvement of joint adhesion (69. 17 ~ 5. 13)o,the treatment group of model of steoarthritis (87. 08 + 5. 32)o, and the difference between the two groups was statistically significant(P 〈0. 05) ; Former Pelletier score was (2. 83 +0. 30), and the latter Pelletier score was (2. 75 -+0. 33), with no statistically sig- nificant ( P 〉0. 05 ) ; the former significantly reduced the number of chondrocytes, matrix stained lighter , Manking score (6. 91±0. 53 ), and the latter increased in the quantity of chondrocytes, chondrocytes ovoid and resumed normal tide line structure, Mankin s score (3. 67 ± 0. 47), and the difference between the two groups was statistically significant (P 〈 0. 01 ) ; Expression of collagen type II positive per- centage comparison, the former(32. 05 ~ 13. 78)% ,the latter (44. 45 + 12. 62)%, and the difference between the two groups was statistically significant (P 〈0. 01 ). Conclusion:AEAB can improve joint adhesions, effectively stimulate proliferation of chondrocyte, increase expression of collagen type II of cartilage, restoring cartilage matrix components and structure of stability tide line, so that repair cartilage damage.
