摘要
目的:研究左旋多巴晶体分子的绝对构型和溶解性特征。方法:采用单晶X射线衍射分析(SXRD)和旋光分析技术考察左旋多巴晶体的手性分子立体空间结构和分子绝对构型,通过理论计算获得晶体样品的晶型纯度,为采用差示扫描(DSC)、红外光谱(IR)分析技术对晶体物质表征提供物质状态依据;通过选择不同pH的溶媒系统研究左旋多巴晶体样品的溶解性质。结果:获得了左旋多巴的分子绝对构型与溶解性特征。结论:掌握了左旋多巴晶体物质的溶解性质,在pH为1.2的0.1 mol·L^(-1)盐酸溶液中晶体样品的溶解性最好,十二烷基硫酸钠(SDS)有明显的增溶作用,该研究为左旋多巴的应用提供基础科学研究数据。
Objective: To study the absolute configuration and the dissolution properties of levodopa. Method: The crystal struc- ture and the absolute configuration of levodopa were confirmed by SXRD and polarimetric analysis, and the crystalline purity was certi- fied by DSC and IR analysis. The dissolution properties of levodopa were studied by the solubility determination in different pH solu- tions. Result: The absolute configuration and the dissolution properties of levodopa were confirmed. Conclusion: The promising disso- lution conditions of levodopa are as follows : 0.1 mol L - 1 hydrochloric acid with pH of 1.2 as the best solution and SDS as the solubi- lizer, which provide the basis for the application research of levodopa .
