酸性染料比色法测定支扩养阴颗粒中总生物碱的含量

Determination of Total Alkaloids in Zhikuo Yangyin Granules by Acidic Dye Colorimetry

目的:探索支扩养阴颗粒中总生物碱的含量测定方法。方法:以盐酸麻黄碱作为参比,通过正交试验确定试验条件,采用酸性染料比色法测定总生物碱。结果:以0.05%溴甲酚绿溶液3 ml为染料,pH 4.2枸橼酸-磷酸氢二钠缓冲液5 ml,三氯甲烷10 ml为萃取溶剂,测定波长为416 nm,盐酸麻黄碱在46～247μg范围内线性关系良好(r=0.999 9),平均加样回收率为98.96%(RSD=0.81%,n=9)。结论:该方法简便可行,可用于支扩养阴颗粒的质量控制。

Objective： To establish a determination method for the total alkaloids in Zhikuo Yangyin granules. Method： Thte total alkaloids were determined by acidic dye colorimetry using ephedrine hydrochloride as the reference substance. The experimental condi- tions were optimized by orthogonal experiment. Result： Bromocresol green liquid （0.05% , 3.0 ml） was used as the colorant, citric acid-disodium hydrogen phosphate buffer（5.0 ml, pH 4.2） was used as the buffer solution, chloroform （10 ml） was applied as the extraction solvent, and the detection wavelength was 416 nm. A good linear range was shown within the range of 46-247 μg （r = 0. 999 9）. The average recovery was 98.96% and RSD was 0.81% （n =9）. Conclusion： The established method is simple and feasi- ble. It can be used in the quality control of Zhikuo Yangyin granules.
