摘要
随机扩增多态性DNA (RAPD)是一种新的分子标记技术 利用RAPD标记对芥菜 (BrassicajunceaCoss .) 16个变种的遗传多样性进行了分析 从 60个 10bp随机引物中筛选出 2 7个有效引物 这 2 7个有效引物共扩增出 336条DNA带 ,其中 2 75条为多态性带 ,占总数的 81 85% 平均每个引物扩增出的DNA带数为 12 4 4 不同引物扩增出各自不同的DNA指纹图谱 ,大部分图谱均有特征或特征带型 芥菜的RAPD多态性百分率和平均每个引物扩增的DNA带数均明显高于已研究的其它物种 。
Random amplified polymorphic DNA (RAPD) is a newly developed molecular marker technology. In this paper, RAPD markers were applied for analysis of the genetic variations among 16 mustard (Brassica juncea Coss.) varieties. 27 10 bp arbitrary primers were screened from 60. 336 DNA bands were amplified by the 27 primers, and among them, 275 were polymorphic ones, which accounted to 81.85% of the total. The average DNA bands amplified by each primer was 12.44. Genomic DNA fingerprints amplified by different primers were obtained. Most fingerprints had their characteristic bands or band patterns. The polymorphic percentage and the average DNA bands amplified by each primer were much higher than those of most studied species, which could be considered as the molecular foundation of rich genetic diversity in mustard.
