Effects of Dehydroandrographolide on levels of blood glucose and expressions of tumor necrosis factor-α in diabetes mellitus rats

摘要 目的观察14-脱羟-11,12-二脱氢穿心莲内酯对链脲佐菌素(streptozotocin,STZ)所致的糖尿病(diabetesmellitus,DM)大鼠血糖及肿瘤坏死因子-α(TNF-α)的影响。方法采用腹腔注射STZ建立DM模型,将成模大鼠随机分为模型组、低剂量组、高剂量组,并设正常对照组。给药14 d后,断尾采血测血糖,苏木精和伊红(HE)染色观察胰岛的形态,免疫组织化学检测TNF-α的表达。结果与正常组比较,模型组血糖明显升高(P<0.05),胰腺组织病理损害严重,TNF-α表达呈强阳性;与模型组比较,低、高剂量组血糖均下降(P<0.05),胰腺组织病理损害减轻,TNF-α表达下调。结论糖尿病时胰岛β细胞中的TNF-α表达增高,14-脱羟-11,12-二脱氢穿心莲内酯可降低血糖,下调TNF-α的表达,减轻胰腺的病理损伤,改善胰岛功能。 Objective To study the effects of Dehydroandrographolide on the levels of blood glucose and expressions of TNF-α in pancreatic tissues of rats with diabetes mellitus (DM) caused by streptozotocin. Methods The DM model rats induced by injecting streptozotocin were randomly divided into the model group, the low dose group and the high dose group, then the normal control group was also set up. The medication was given for 14 days. The fasting blood glucose was detected. The structural changes of pancreatic tissues stained with hematoxylin and eosin were observed under a microscope. The expressions of TNF-α were analyzed by immunohistochemistry. Results Compared with the normal control group, the levels of blood glucose obviously increased in the model group (P 〈 0.05), the pathological injuries of the pancreatic tissues were serious, and the expressions of TNF-α were shown to be strongly positive. Compared with the model group, the levels of blood glucose decreased in both the low dose group and high dose group (P 〈 0.05), with alleviated pathological injuries of the pancreatic tissues, and the expressions of TNF-α in both groups decreased. Conclusion The expression of TNF-α in pancreatic 13 cell increased when suffered from diabetes mellitus. Dehydroandrographolide can reduce the level of blood glucose, down-regulate the expression of TNF-α, lessen the pathological injuries of pancreatic tissues, thus ameliorate the functions of pancreatic islet.
