摘要
目的分析H1-钙结合蛋白(H1-calponin)在胃癌组织中的表达,探讨其与基质金属蛋白酶2(MMP-2)表达的关系及临床意义。方法应用反转录-聚合酶链反应(RT-PCR)及Western印迹分析检测胃癌组织(n=20)及癌旁正常组织中H1-calponin的mRNA和蛋白表达水平;RT-PCR和明胶酶图检测胃癌组织及癌旁正常组织中MMP-2的mRNA水平及酶活性。结果 Western印迹灰度扫描显示,胃癌组织中H1-calponin表达水平显著低于癌旁正常组织(0.98±0.28vs 3.11±1.37,t=-6.833,P<0.01),而MMP-2的活性显著高于癌旁正常组织(3.48±1.25vs 1.26±0.39,t=7.556,P<0.01),胃癌组织中H1-calponin表达水平与MMP-2活性呈负相关(r=-0.623,P<0.01)。结论 H1-calponin在胃癌组织中表达下调且与MMP-2活性呈负相关,表明其在胃癌发生过程中发挥重要作用。
Objective To investigate the expression level of H1-calponin and analyze its relationship with matrix metalloproteinase 2(MMP-2) expression level as well as their clinical significance in gastric cancer. Methods Reverse transcription-polymerase chain reaction(RT-PCR) and Western blotting were performed to detect H1-calponin mRNA and protein levels in gastric cancer( n = 20) or adjacent normal gastric mucosa tissues. Zymography and RT-PCR were used to detect the activity and mRNA levels of MMP-2, respectively,in gastric cancer or adjacent normal gastric mucosa tissues. Results In gray scale scanning with Western blotting, H1-calponin gene expression of gastric cancer was significantly lower than that of adjacent normal gastric mucosa tissues (0.98± 0.28 vs 3.11 ±1.37, t =- 6. 833, P 0.01). MMP-2 activity in gastric cancer cases was significantly higher than that in adjacent normal gastric mucosa tissues (3.48±1.25 vs 1.26±0.39, t = 7. 556, P 〈0.01). A negative correlation was found between H1-calponin protein level and MMP-2 activity in gastric cancer tissues (r = -0. 623, P〈 0.01 ). Conclusion H1-catponin expression level is decreased in gastric cancer,which suggested the important role of H1-calponin in gastric cancer.
