多头香石竹“莱拉克”的组培快繁技术初探

摘要 以多头香石竹"莱拉克"的茎段为外植体,进行组织培养快速繁殖,探讨不同培养基对香石竹诱芽萌动、丛芽增殖、生根培养的影响。结果表明:①当培养基为MS+6-BA1.0 mg/L+0.08%琼脂+0.30%蔗糖时,适于芽的诱导;②当培养基为MS+6-BA0.8 mg/L+NAA0.2mg/L+0.08%琼脂+0.30%蔗糖时,继代增殖效果最好;③当培养基为1/2MS+NAA0.8 mg/L+0.10%活性炭+0.15%蔗糖时,生根效果最好,有效生根率达95.6%;④调整激素比例,适当降低细胞分裂素并微量增加生长素的浓度,可减少组培苗玻璃化的程度;⑤pH偏低,基质较软的培养基会增加苗的玻璃化程度;⑥0.10%微量活性炭的使用,不仅可以减少玻璃化的产生,还可以促进"莱拉克"的生根。 Taking the stem segments of Carnation Lilac as explants, we conducted the study of tissue culture and rapid propagation to discuss the influences of different culture medium to induced bud germination, bud proliferation and rooting culture. The results were as follows. The best bud induction medium was MS with 6-BA1.0 rag/L, 0.08% agar and 0.30% sucrose. The optimal media for subculture multiplication were MS media supplemented with 6-BA0. 8 mg/L, NAA 0. 2 mg/L ,0. 08% agar and 0. 30% sucrose , while the best medium for rooting was 1/2 MS with NAA 0.8 mg/L, 0.10% activated carbon, 0. 08% agar and 0. 15% sucrose , and the rooting rate could reach up to 95.6%. In addition, it could decrease the degree of vitrification of tissue culture seedlings by adjusting hormone ratio, reducing the cytokinin properly and increasing little amount of the concentration of ghrelin. What's more, the medium with low pH and softer matrix would increase the vitrification of seedlings. Finally, it couldn't only reduce the degree of vitrification, but also promote rooting of the bull carnation Lilac by use of 0.10% micro activated carbon,.
