摘要
目的研究美国肉参中分离得到的岩藻聚糖硫酸酯(SC-FUC)对化学缺氧条件下人胃癌HGC-27细胞转移作用的影响。方法利用化学法建立HGC-27细胞体外缺氧培养模型。采用MTT法检测了SC-FUC对HGC-27细胞增殖活性的影响,细胞粘附实验研究了SC-FUC对肿瘤细胞粘附能力的影响,Transwell小室法观察了对HGC-27细胞迁移和侵袭能力的影响,体外小管形成和鸡胚尿囊膜实验(chorioallantoic membrane assay,CAM)观察了SC-FUC抑制血管新生的情况。结果 SC-FUC具有显著抑制HGC-27细胞增殖的活性(72和96 h的IC50值分别为170和100 g/ml);可降低肿瘤细胞同基质和血管内皮细胞间的粘附率,并抑制HGC-27细胞的侵袭和迁移能力,其72h的侵袭率和迁移率分别降低了59.73%和67.96%(P<0.05,P<0.01);能显著抑制体外内皮细胞小管形成能力,并减少体内CAM新生血管数目。结论 SC-FUC具有显著抑制缺氧诱导的HGC-27胃癌细胞转移的能力。
Objective To investigate the anti-metastatic effects of sea cucumber fucoidan (SC-FUC) extracted from lsostichopus badionotus on human gastric carcinoma cell line HGC-27 under hypoxia induced by COC12. Method The chemical hypoxia was induced by 25 ~tmol/L COC12. The cell proliferation of HGC-27 cell was determined by MTT assay. Cell adhesion assay and transwell assay were applied to detect the anti-tumor metastasis in HGC-27 cell, In addition, in vitro tubule formation assay was used to demonstrate the effect of angiogenic ability on endothelial cells and chick chorioallantoic membrane (CAM) model was used to check whether the neovascularization of CAM could be suppressed in vivo. Results SC-FUC could suppress the proliferation of HGC-27 cells under cobalt-induced hypoxia (P〈0.05, P〈0.01). Moreover, the adhesion with Matrigel and HUVEC cells, migration and invasion of HGC-27 cells were inhibited. SC-FUC significantly inhibited the tube formation in vitro and decreased the CAM in the number, diameter and thinner, distribution messy. Conclusion SC-FUC has anti-metastatic and angiogenesis effects on HGC-27 cells under hypoxia.
