摘要
目的:观察槲皮素对糖尿病大鼠超氧化物歧化酶(SOD),丙二醛(MDA),一氧化氮(NO)及一氧化氮合酶(NOS)的影响,分析槲皮素对糖尿病大鼠的肾脏保护机制。方法:采用一次性腹腔注射链脲佐菌素建立糖尿病模型。将大鼠随机分为3大组,每组再设2个小组(n=8):正常组(A1,A2),糖尿病模型组(B1,B2),槲皮素治疗组(C1,C2),分别于槲皮素(50 mg.kg-1.d-1)ig治疗6周和12周后处死取双肾,计算各组大鼠肾脏肥大指数;一肾用苏木精-伊红染色,光镜下观察肾小球病理形态学变化;另一肾测定其内SOD活性、MDA、NO含量及NOS活力的变化。结果:槲皮素治疗12周肾脏肥大指数(6.99±0.58)×10-3,与模型组(8.92±0.66)×10-3相比明显降低(P<0.05)。槲皮素治疗6周和12周肾组织SOD活性(165.31±14.89),(148.33±13.22)U.mL-1,与模型组(149.02±17.21),(121.32±15.33)U.mL-1相比明显升高(P<0.05);MDA含量(17.89±2.88),(19.46±3.02)μmol.L-1,NOS活性(15.13±2.89),(17.65±2.46)ng.L-1,NO含量(0.42±0.12),(1.41±0.48)μmol.L-1,与模型组MDA含量(19.68±3.01),(21.12±2.99)μmol.L-1,NOS活性(18.23±2.42),(21.22±3.01)ng.L-1,NO含量(0.73±0.25),(1.91±0.35)μmol.L-1相比明显下降(P<0.01);且明显改善肾小球病理变化。结论:槲皮素可通过增加氧自由基的清除,抑制NOS的活性及NO的含量保护糖尿病肾脏。
Objective:To observe the effect of quercetin on free radical and nitric oxide (NO) and nitric oxide synthase (NOS) change after streptozotocin-induced diabetic in rats and analyze the protectiive mechanisms of quercetin on kindey in diabete rat. Method: The diabetic rats were induced by a single intraperitoneal injection of streptozotocin. The rats were randomly divided into three groups:normal group(A1, A2), diabetic model group(B1,B2), quercetin treatment group (C1, C2), and each group was set into two groups (n=8). Respectively they were applied for the quercetin (50 mg·kg-1·d-1) lavage treatment for 6 weeks and 12 weeks and then killed for kidneys. The cell morphologic changes of renal tissue were observed by hematoxylin and eosin staining. The rats' renal hypertrophy index was calculated. Changes of the activity of superoxide dismutase (SOD) and NOS, the content of malondialdehyde(MDA)and NO in renal tissue were observed by means of spectro-photometry. Result: Quercetin treatment for 12 weeks could obviously reduce renal hypertrophy index (6.99±0.58)×10-3,(P〈0.05).Quercetin treatment for 6 weeks and 12 weeks could obviously enhance SOD activities(165.31±14.89),(148.33±13.22)U·mL-1,P〈0.05), reduce the MDA conten (17.89±2.88),(19.46±3.02)μmol·L-1,(P〈0.01), the activities of NOS(15.13±2.89),(17.65±2.46)ng·L-1,(P〈0.01)and the contents of NO(0.42±0.12),(1.41±0.48)μmol·L-1,(P〈0.01), and obviously improve the glomerular pathological changes. Conclusion: Quercetin can protect diabetic kidney of the rats with quercetin, which may be mediated by its anti-free radical and reducing the contents of NO and the activities of NOS.
