摘要
目的:探讨胎盘转化生长因子-β(TGF-β)三种亚型及其受体的表达与妊娠期肝内胆汁淤积症(ICP)的关系。方法:用免疫组织化学染色技术(SP法)检测ICP患者和正常妊娠胎盘组织中TGF-β1、2、3三种亚型和两种功能型受体(TGF-βRⅠ和TGF-βRⅡ)的表达水平。结果:ICP患者胎盘绒毛TGF-β2表达的平均面积、染色强度和IOD值均明显小于正常妊娠组(P<0.01),其IOD值与母血TBA呈负相关(r=-0.824,P<0.01);TGF-βRⅡ的表达面积和IOD值高于正常妊娠组(P<0.01,P<0.05);而TGF-β1、3和TGF-βRⅠ与正常相似。结论 :ICP患者存在免疫耐受-免疫排斥不平衡的原因可能与TGF-β2/TGF-βRⅡ表达异常有关。
Objective: To explore the relationship between expressions of three transforming growth factor -β( TGF-β) subtypes and the receptors and intrahepatic cholestasis of pregnancy (ICP) . Methods: Immunohistochemical SP method was used to detect the expression levels of TGF-β1, TGF -β2, TGF -β3, TGF-β R I , and TGF-β R lI in placental tissues of ICP patients and normal pregnant women. Results : The mean area, staining intensity, and IOD value of TGF-β2 in placental villi of ICP group were statistically significantly lower than those in normal pregnancy group (P 〈 0. 01 ) , there was a negative correlation between IOD value and maternal blood total bile acid (TBA) (r = - 0. 824, P 〈 0. 01 ) ; the area and IOD value of TGF -β R II in ICP group were statistically significantly higher than those in normal pregnancy group (P 〈 0. 01, P 〈 0. 05 ) ; but there was no statistically significant difference in TGF -β1, TGF -β3, and TGF-β R I between the two groups. Conclusion : The cause of imbalance of immune tolerance - immune rejection in ICP patients may be correlated with abnormal expressions of TGF-β2/TGF-β R II.
