响应面法优化红雪茶中类胡萝卜素的提取工艺 被引量:5

摘要 为确定红雪茶类胡萝卜素的最佳提取工艺条件,在单因素实验的基础上,选择丙酮为浸提溶剂,料液比、提取温度、提取时间作为自变量,以类胡萝卜素特定波长下吸光度值作为响应值,基于Box-Behnken中心组合方法进行三因素三水平的实验设计,根据响应面结果,建立了回归模型。研究结果表明,红雪茶类胡萝卜素的最优提取工艺条件参数为:料液比1∶3(g/mL),提取温度52℃,提取时间20min。在该条件下,红雪茶类胡萝卜素提取液的吸光值为0.3682,实际获取红雪茶类胡萝卜素量22.092μg/g,与预测值相对误差较小。说明该模型拟合度好,优化后得到的提取条件准确、可靠。 Based on Box-Behnken center composite design within Design-Expert7.0 software and response surface results,the optimal extraction process for carotenoids from Lethariella spp.were determined and a regression model was established.On the basis of single factor tests,using absorbance values in a specific wavelength of carotenoids as response value, three key factors (solid- liquid ratio, extraction temperature and time)were selected as independent variables and acetone as extraction solvent.The results showed that the optimal extraction process of carotenoids from Lethariella spp.was obtained as followed.solid-liquid ratio of 1:3 (g/mL) ,extraction temperature of 52℃,and extraction time of 20min. Under these conditions, the extraction of carotenoids from Lethariella spp. absorbance was 0.3682, i.e.22.092μg from per g Lethariella spp. raw materials.The results showed that regression equation fit well with experimental data because of a good accordance between the predicted and observe values. Meanwhile the optimized technology was stable and viable with minor error and good degree.
