摘要
利用超声提取法对云南省不同地区不同品种辣椒样品进行预处理,采用高效液相色谱法(HPLC)测定样品中辣椒碱和二氢辣椒碱的含量。具体方法为:采用色谱柱HanbonSci&Tech.LichrospherC18(250×4.6mm,5μm)分离,甲醇-3‰磷酸水溶液梯度洗脱(65%~90%,0~30min),检测波长280nm,辣椒碱和二氢辣椒碱的相对标准偏差都小于1%,平均回收率都在99%以上。该方法具有较好的分离效果和较高的稳定性,适用于云南省不同地区不同品种中辣椒碱和二氢辣椒碱含量的测定。测定结果显示云南省不同地区不同品种不同成熟度的辣椒中辣椒碱和二氢辣椒碱含量差异很大。
A high-performance liquid chromatograph (HPLC)method was developed for determinating contents of capsaicine and dihydrocapsaicin in different kinds chills in Yunnan Province.Chills was pretreated by the ultrasonic extraction,and capsaicine and dihydrocapsaicin were separated and assayed with reversed phase C18 column. Finally the determination was carried out using HPLC gradient elution method, using methanol-3%o phosphate as the mobile phase(65% -90% ,0-30min) at a flow rate of 1.2mL/min.The detector wavelength was set at 280nm and the injection volume used was 20pL. The relative standard deviations of capsaicine and dihydrocapsaicin determinated were all less than 1% and the average recoveries was all above 99%.This method showed good separation effect and high stability, and could be used for determining the content of capsaicine and dihydrocapsaicin in different kinds of chills in Yunnan Province.The result demonstrated that the contents varied largely from different kinds, different degrees of maturity and different districts.
