摘要 采用MTT法检测玉米芯水提物对人结肠癌细胞株SW480、SW620、DLD1以及正常人肝细胞HL-7702的增殖活力的影响;倒置显微镜下观察细胞形态的变化;DAPI染色法检测该水提物对不同细胞株细胞核凋亡的影响。结果表明:不同浓度(1、2、4、6 mg/mL)的玉米芯水提物对人结肠癌细胞株均有抑制能力,且呈明显的量效依赖关系。用该水提物处理正常细胞HL-7702,在低浓度时没有抑制作用,在高浓度时略有抑制作用。该水提物对各细胞株作用24 h,半数抑制浓度(IC50)分别为2、3、4、6 mg/mL。显微镜观察表明:经该玉米芯水提物处理后的结肠癌细胞,形态发生明显的改变,细胞形状变圆,贴壁细胞数量明显减少。DAPI染色结果显示:多数细胞核表现出染色质的不均一性,有典型的"梅花"状凋亡小体出现。 The growth inhibition and apoptosis effects of aqueous extract of corncob on colonic-cancer cell strains SW480,SW620,DLD1 as well as normal human hepatic cell 7702 in vitro were studied by MTF assay and DAPI staining. Morphological changes were observed by inverted microscope. The results showed that the water etract of corncob with the different concentrations (1,2,4,6 mg/mL) suppressed the proliferation of different colonic-cancer cell strains, and displayed significantly in a dose-dependent manner. The ICso values were 2,3,4,6 mg/mL for 24 h treatment. The microscopic observation showed that cellular morphology treated by the aqueous extract obvious changed. The attached cells were largely reduced and getting round. Meanwhile, the cells were detached in large areas from culture dishes. The analysis by DAPI staining indicated that many nuclei showed the uneven chromatin distribution in a typical apoptotic body shape of "the plum blossom".
