摘要
目的研究去整合素对后发障形成的影响,探讨整合素β1在HLECs转分化中的作用机制。方法免疫荧光染色检测整合素βl在后发障动物模型中的表达、逆转录聚合酶链反应(RT—PCR)检测整合素BlmRNA的变化、酶联免疫法检测房水中TGF-132的变化。免疫荧光染色检测整合素βl在后发障体外模型中的表达,逆转录聚合酶链反应(RT—PCR)检测整合素plmRNA的变化,去整合素(Kistrin)进行干预后检测整合素βl及其mRNA的表达。结果在兔的后发障动物模型及离体后发障模型中,整合素βl及其mRNA的表达在术后6周表达最强,在术后8周表达减弱。整合素B1及其mRNA在术后6周的表达与术后4周、8周相比,差异有显著性(P〈0.05)。在离体后发障动物模型中,使用去整合素(Kistrin)进行干预后,干预组整合素β1表达免疫荧光面积为(0.03±0.02)%,对照组为(0.14±0.06)%,差异有显著性(P〈0.05);模型中整合素βl及其mRNA的表达下降(P〈0.05)。结论整合素β1参与了后发障的发生,可以通过抑制整合素B1的表达来抑制后发障的发生。
Objective To study the effect of disintegrins on posterior capsular opacity (PCO), and explore the mechanism of integrin β1 in transdifferentiation of human lens epithelial cells (HLECs). Methods We used immunofluorescence technique to detect the expression of integrin- β 1 during after-cataract, setting up a model of PCO in vitro consisted, to research LECs'proliferation, and using reverse transcription-polymerase chain reac- tion (RT-PCR) to detect changes of integrin- β 1 mRNA. ELISA was used to detect TGF- β 2 changes to view the influence of them during PCO. We set up a model of PCO by New Zealand white rabbit, observed PCO oc- currence and used immunofluorescence technique and RT-PCR to detect the expression of integrin- β 1 during after-cataract. At the same time viewed the change of TGF-β2 in aqueous humor. We also set up a vitro model of PCO and used immunofluorescence technique and RT-PCR to detect the expression of integrin- β 1 during after-cataract. Results Peplos' becoming turbidity started in 4 weeks later after operation;6 weeks and 8 weeks after operation, turbidity turned heavier. At 6 weeks after operation, the expression of integrin- β 1 in pe- plos was most, and then with reduction, the same with TGF- 13 2 change. With the extension of time, all the ex- pression of cytokine had declined. Conclusion Integrin can effectively inhibit lens epithdial cell migration, slow down the development of PCO.
