摘要
目的应用反相高效液相色谱法(RP-HPLC)测定胎盘中的新洁尔灭。采用甲醇浸泡溶解,10%高氯酸沉淀蛋白的方法,提取样品后进行高效液相色谱测定。方法色谱柱:用Acclaim C18色谱柱(250mm×4.6mm i.d.,5μm);流动相:甲醇-乙腈-水(53:20:27);紫外检测器:检测波长:254nm;柱温:30℃;流速:0.85mL/min。结果在2.0~10.0μg·mL-1质量浓度范围内,呈现良好线性关系(r:0.9999),回收率为95.7%~99.6%,日内RSD为1.4%~1.8%,日间RSD为1.5%~2.1%。结论该方法简便、快速、灵敏、准确,完全符合中国药典2010版规定,可作为临床检测方法。
Objective Apply the HPLC to determine Bromogeramine in the placenta. Use methanol to dissolve andl0% Perchloric acid to subside protein to get the sample which is tested by HPLC. Methods Column: Acclaim C18 (250mm×4.6mm i.d.,5μm) mobile phase methanol: Methanol: Acetonitrile: water (53:20:27) wave length:254nm column temperature 30℃ flow rate: 0.85 ml/ min. Results The during concentration range 2.0-10.0 μg·mL-1 shows a good linear relationship (r:0.9998). recovery rate is 95.7% -99.6%, Intra-day RSD isl.4%-1.8%, Inter-day RSD is 1.5% - 2.1%. Conclusion The method is simple, rapid, sensitive, accu- rate and fully complies with the provisions of the Chinese Pharmacopoeia 2005 edition, which can be used as clinical testing method.
