摘要
目的建立一种测定睾丸组织中头孢匹胺的RP-HPLC检测方法,用于头孢匹胺在小鼠睾丸组织中的分布和消除规律研究。方法睾丸组织匀浆液样品经液-液萃取后,以甲醇-三乙胺醋酸-水=22-0.2-77.8(v/v)为流动相,流速1.0mL.min-1,采用大连依利特BDS柱(250mm×4.6mm;i.d.,5μm)色谱柱分离,柱温30℃,紫外检测波长254nm。结果标准曲线线性范围0.1-50mg.kg-1,线性关系良好(r>0.999),定量限为0.1mg.kg-1,回收率95%以上,日内和日间变异系数均低于10%。结论该方法操作简便、灵敏、准确,适合于临床前药动学及组织分布的初步研究。
OBJECTIVE To develop an HPLC-UV method for the determination of cefpiramide and detect the distribution and elimination of cefpiramide in rat testis tissue. METHODS After a simple liquid-liquid extraction, cefpiramide were separated on elite BDS C18 column(4. 6mm × 250mm,5 μm) and detected by UV spectrometry. The detection wavelength was set at 254nm. The mobile phase consisted of methanol-triethylamine acetic acid-water (22- 0. 2-77. 8) at a flow rate of 1.0mL .min^- 1. The column temperature was set at 35℃. RESULTS The linear cali- bration curves were obtained in the concentration range of 0. 5 -50. 0mg.kg^- 1 ( r 〉 0. 999 ). The lower limit of quantification was 0. 1mg . kg^-1. The recoveries were more than 95.0%, and the intra-and inter-day coefficients of variation were less than 10% ( RSD 〈 10% ). CONCLUSION The RP-HPLC method is specific, simple, sensitive and accurate. And can be used for pilot study on quantitate cefpiramide in rat testis tissue.
