摘要
目的 :探讨内皮依赖性舒张因子 (EDRF/NO)对豚鼠耳蜗微循环的调节作用。方法 :2 8只豚鼠 ,随即分为4组 ,每组 7只。生理盐水组静脉注射生理盐水 ,L Arg/S组和L Arg/L组静脉注射不同剂量L 精氨酸 ,L NNA组静脉注射L 硝基 精氨酸。行耳蜗开窗后 ,利用计算机图像分析系统 ,对豚鼠耳蜗的血流、血管管径、血压、心率、血气进行分析。观察了不同剂量L 精氨酸 (L Arg)及L 硝基 精氨酸 (L NNA)对豚鼠耳蜗血流 (CoBF)的作用。结果 :L Arg/S组 ,用药后耳蜗血液流速加快 ,管径扩张 ,平均动脉压均下降 ;L Arg/L组 ,用药后耳蜗血流速度增加后流速减慢 ,管径扩张 ,血压升高 ;L NNA组 ,用药后流速减慢 ,血管管径收缩 ,但持续时间很短。与用药前相比差异均有显著性 (P <0 .0 5 )。结论 :EDRF/NO对CoBF有调节作用 ,适当剂量的EDRF/NO可扩张血管 ,加快血流 。
Aim: To investigate the role of endothelium derived relaxing factor(EDRF/NO)in regulating cochlear microcirculation.Methods: The total of 28 guinea pigs were randomly divided into 4 groups (7 guinea pigs each group). Normal saline group was injected with normal saline. L Arg/S group and L Arg/L group were injected with L Arg. L NNA group was injected with L NNA. After cochlear fenestration was done with intravital microscopic examination, cochlear blood flow(CoBF),vessel diameter,blood pressure of guinea pigs were analysed using image processing technique.Results: In L Arg/S group CoBF increased, micro vessel was dilated, and mean arterial pressure declined;In L Arg/L group CoBF increased,and slackened later, micro vessel was dilated, mean arterial pressure was elevated; In L NNA group CoBF slackened ,micro vessel contracted. In all 3 groups there were significant difference compared with those before administration( P <0.05). Conclusion: CoBF can be regulated by EDRF/NO in guinea pigs. EDRF/NO can be used to treat inner ear disorder by dialating vessel and increasing blood velocity.
