小柴胡汤含药血清诱导SSMC-7721人肝癌细胞株分化研究

Study on Differentiation of Human SMMC-7721 Cells by Medicated Serum of Xiaochaihu Decoction in Vitro

目的:观察小柴胡汤含药血清对SMMC-7721人肝癌细胞是否具有分化诱导作用,并探讨其分化诱导的作用机制。方法:经药物作用后,观察细胞形态、MTT抑制率、乳酸脱氢酶(LDH)、琥珀酸脱氢酶(SDH)、细胞周期的情况。结果:小柴胡汤含药血清使SSMC-7721细胞株有向成熟细胞形态分化的趋势;MTT抑制作用增强;细胞内LDH活性降低、SDH活性上升;流式测定S期细胞数减少,细胞周期阻滞于G0/G1期。其中以低剂量组效果最为显著(P<0.05)。结论:小柴胡汤含药血清可诱导SSMC-7721细胞株向成熟细胞分化,其作用机制可能与降低细胞内LDH活性、升高SDH活性,并且将细胞阻滞于G0/G1期有关。

Objective： To observe the effects of SMMC-7721 cell differentiation induced by medicated serum of Xiaochaihu decoction and to discuss its mechanism. Methods ： The cells differentiation was observed on some changes of eytomorphology, including inhibiting rate in MTT, lactate dehydrogenase （ LDH ）, succinate dehydrogenase （ SDH ）, and cell cycle. Results ： Compared with the control group, the SMMC-7721 cell differentiation can induced mature cell morphologically, enhance inhibition of MTT, reduce LDH activities and increase SDH activities in cell. Compared with the control group, SMMC-7721 cell cycle was blocked into G0/Glphase and the cell amount in S phase was decreased. And that of low- dose Xiaochaihu decoction group was significant （ P〈0.05 ） .Conclusion ： SMMC-7721 cells can be induced differentiation intomature granulocyte by medicated serum of Xiaochaihu decoction.
