摘要
目的研究束缚应激条件下大鼠体重、行为及生化指标变化。方法选择Wistar大鼠30只随机分为3组:正常对照组,束缚3周组和束缚5周组。监控大鼠体重变化,并采用酶联免疫吸附测定法(ELISA)检测束缚应激大鼠血清皮质酮(CORT)、去甲肾上腺素(NE)、热休克蛋白70(HSP70)、IL-2和IL-4的含量变化。结果束缚应激后,大鼠体重增长缓慢且有明显的行为变化,5种生化因子含量也均有明显的变化。与对照组相比,束缚3周和5周之后,CORT、NE、HSP70、IL-4含量分别升高了11.25%和46.65%、25.16%和39.09%、5.15%和13.49%、9.86%和18.69%(除束缚3周组HSP70外,其余均P<0.05),且NE、CORT和IL-4含量升高具有时间依赖性;IL-2含量显著降低了15.43%和16.01%(P<0.05),但不具有时间依赖性。结论束缚应激可改变大鼠免疫功能及调节神经递质作用,CORT、NE、HSP70、IL-2和IL-4可作为重要的分子靶标。
Objective To investigate the effects of restraint stress on body weight, behaviour and biochemical index of model rats. Methods The content changes of CORT, NE, HSPT0, IL - 2 and IL - 4 in serum of restraint stress rats were measured by enzyme -linked immunosorbent assay (ELISA). Results The results indicated that restraint stress could sup- press the weight increasing and change the behaviour of the rats. The contents of CORT, NE, HSP70 and IL - 4 increased significantly by 11.25% and 46.65% ,25.16% and 39.09% , 5.15% and 13.49% ,9.86% and 18.69% in time - de- pendent manner except for HSPT0 after restraint stress 3 and 5 weeks compared to control, while the contents of IL - 2 de- creased 15.43% and 16.01% after restraint stress hut not time - dependent. Conclusion Restraint stress could change the immunity function and regulate neurotransmitter, CORT, NE, HSP70, IL - 2 and IL - 4 might be used as crucial molecular markers.
