银屑病患者外周血T细胞Notch信号通路相关基因表达的研究 被引量:9

Expression of genes involved in the Notch signaling pathway in peripheral blood T cells from patients with psoriasis

摘要 目的分析寻常性银屑病患者外周血T细胞Notch信号通路及其下游靶基因Hes-1表达的情况。方法取20例银屑病患者及20例健康人外周血,分离、培养、扩增外周血T细胞并鉴定纯度;提取纯化mRNA并反转录合成cDNA,实时荧光定量PCR(RFQ—PCR)检测Notchl、Notch2及Hes-1在mRNA水平表达。结果银屑病患者外周血T细胞Notchl、Notch2受体mRNA表达水平是对照组的1.8391、1.8423倍(2值);Notch信号通路的靶基因Hes-1在银屑病外周血T细胞表达水平2值为2.5123。结论Notch信号通路的高表达可能参与了银屑病患者外周血T细胞的活化,与银屑病发病具有密切关系。 Objective To detect the expression of the Notch signaling pathway, and a downstream targetgene of the pathway, Hes-l, in peripheral blood T cells of patients with psoriasis vulgaris. Methods Peripheral blood samples were collected from patients with psoriasis vulgaris and healthy human controls, followed by the isolation, culture, amplification and purification of peripheral blood T cells. Then, mRNA was extracted from the T cells and reversed to cDNA. Real time fluorescent quantitative PCR (RFQ-PCR) was performed to determine the expressions of Notch 1, Notch 2 and Hes-1 genes. Results The expression intensity of Notch 1, Notch 2 and Hes-1 genes in peripheral blood T cells from patients with psoriasis was 1.8391, 1.8423 and 2.5123 times that from the healthy human controls, respectively. Conclusions The overexpression of genes involved in the Notch signaling pathway may participate in the activation of peripheral blood T cells, which appears to be closely associated with the pathogenesis of psoriasis.
