摘要
为探索适宜莱氏野村菌生长的营养条件,研究了12种碳源、6种氮源、5种金属离子、7种维生素及不同碳氮比对莱氏野村菌Nr16菌落生长速度、菌落直径、菌丝干重及产孢量的影响。结果表明:莱氏野村菌Nr16液体、固体培养对各营养物质的需求并不是完全一致的,不同营养物质对该菌的菌落生长、菌丝干重及产孢量有明显影响。莱氏野村菌Nr16菌落生长的最佳碳源为葡萄糖,最佳氮源为酵母膏,最优碳氮比为20∶1,Zn2+对菌落的生长有利,维生素对菌落的生长无显著影响;液体菌丝生长的最佳碳源为葡萄糖,氮源为酵母膏,碳氮比为10∶1,金属离子对其无显著影响,维生素中VC对其生长有明显促进作用,而VB1与VB2对液体菌丝的生长有明显的抑制作用;明显促进该菌产孢的最佳碳源为葡萄糖,氮源为酵母膏,碳氮比为20∶1,金属离子对产孢无显著影响,其中,Cu2+对产孢有一定的抑制作用,维生素对产孢亦无显著影响,叶酸对该菌的产孢有显著抑制作用。
Twelve carbon sources, six nitrogen sources, five metal ions, seven vitamins and different ratio of carbon to nitrogen were used in N. rileyi culture to study effects on colony growth rate, colony diameter, dry mycelium weight and sporulation quantity of N. rileyi Nr 16. The results showed that there was difference in demand of nutrient substances between liquid culture and solid culture, different nutrient substance had obvious effects on colony growth, dry mycelium weight and sporulation quantity. Glucose, yeast extract and 20 : 1 ratio of C/N were the best for colony growth of N. rileyi Nr 16 but vitamins had no significant effect on colony growth. Glucose, yeast extract, 10 : 1 ratio of C/N and Vcwere beneficial to hyphal growth of Nomuraea rileyi Nr 16 but metal ions had no significant effect on hyphal growth and VB1 and VB2 had obvious inhibition effect on hyphal growth under liquid culture. Glucose, yeast extract and 20 : 1 ratio of C/N could improve sporulation quantity of N. rileyi Nr 16 obviously. Metal ions and vitamins had no significant effect on sporulation quantity but Cu2+ and folic acid had the inhibition and obvious inhibition effect on sporulation quantity.
