摘要
目的:评价U0126对人黑素瘤细胞A375侵袭的抑制作用。方法:人黑素瘤A375细胞在6孔细胞培养板中培养,加U0126处理过夜,然后通过观察A375细胞的细胞形态、细胞移动及细胞侵袭判断U0126对A375细胞的抑制。蛋白电泳试验检测U0126对黑素瘤细胞A375的ERK1/2蛋白磷酸化的影响。结果:U0126诱导A375细胞产生肌动蛋白(actin)张力丝,导致细胞形态改变;A375细胞的移动和细胞侵袭被抑制。结论:U0126可能通过抑制黑素瘤的ERK1/2蛋白的磷酸化从而抑制人黑素瘤细胞A375的侵袭。
Objective: To assess the inhibitory effect of U0126 on the invasion of melanoma cell A375. Meth- otis: Human melanoma cells A375 were cultured in six-well culture plate. U0126 was added to the culture media for over-night as experimental group and DMSO was used as control group. Cell morphology, cell moving and cell inva- sion ability assay were used to assess the inhibitory effect of U0126 on the invasion of melanoma cell A375. Protein electrophoresis method was used to determine the effect of U0126 on the protein phosphorylation of the ERK1/2 of melanoma cell A375. Results: The production of actin tonofilament induced by U0126 resulted in morphological changes of the melanoma cell A375. The moving ability and invasion of the melanoma cell A375 were inhibited. Conclusion: U0126 can inhibit the invasive activity of melanoma cells A375 through inhibiting the protein phospho- rylation of the ERK1/2 of melanoma cell A375.
