摘要
目的探讨膀胱癌中脑恶性肿瘤缺失基因1(DMBT1)的表达及其与肿瘤生物学行为的关系。方法采用RT-PCR和免疫组化SP法检测70例膀胱癌组织和12例正常膀胱组织中DMBT1 mRNA及蛋白的表达。结果膀胱癌组织中DMBT1 mR-NA和蛋白的表达水平低于正常膀胱组织(P<0.05),随着病理分级和临床分期的升高,DMBT1 mRNA和蛋白的表达水平明显下调(P<0.05)。DMBT1在有淋巴结转移的癌组织中表达明显低于在无淋巴结转移癌组织中的表达(P<0.05)。结论 DMBT1基因在膀胱癌组织中存在表达缺失和低表达,提示DMBT1基因在膀胱癌的发生、发展中可能具有抑癌基因的作用。
Purpose To investigate the expression of deleted in malignant brain tumor 1 ( DMBT1 ) in bladder urothelial carcinoma and the relationship to biological behavior of the cancer. Methods The expression of DMBT1 in 70 cases of bladder urothelial carcinoma and 12 specimens of normal bladder tissues was detected by SP method of immunohistocbemistry and semiquantitative RT-PCR. Re- sults The expression of DMBT1 protein and mRNA in bladder cancer was significantly lower than that of normal bladder epithelium. The expression of DMBT1 protein and mRNA was significantly decreased in higher staging and grading tumors. The expression of DMBT1 in lymph node metastasis group was lower than that in lymph node negative group ( P 〈 0. 05 ). Conclusions The expression of DMBT1 gene is lost and reduced in bladder urothelial carcinoma, which indicates that the DMBT1 gene may play an important role in suppressing the tumorigenesis and development in bladder urothelial carcinoma.
