植物生长调节剂对3个大花蕙兰(Cymbidium hybridium)品种类原球茎及不定芽诱导的影响 被引量:3

摘要 研究了多种植物生长调节剂(Plant growth regulator,PGR)对3个大花蕙兰品种‘金色年华'、‘绿洲'和‘华尔兹'类原球茎(Protocorm-like body,PLB)和不定芽诱导的影响。结果表明:PGR在一定浓度下能促进类原球茎的产生,MS培养基上PLB诱导的最适PGR配比为BA 4 mg/L+NAA 0.5 mg/L+KT 0.5 mg/L+TDZ 0.2 mg/L,增殖系数分别为11.12、9.65和7.54,与对照(BA 4 mg/L+NAA 0.5 mg/L)相比差异达到极显著水平。选取PLB诱导率最低的品种‘华尔兹'建立诱导不定芽的离体再生途径,筛选出不定芽诱导的最佳PGR配比为BA 2 mg/L+NAA 0.2 mg/L+PIC(毒莠定,Picloram)1 mg/L,平均增殖系数为5.08。于MS培养基中添加NAA 0.2 mg/L+HMI(恶霉灵,Hymexazol)2 mg/L,4 d时的生根率高达85%。试验表明:大花蕙兰通过类原球茎和不定芽的诱导均能实现产业化繁殖种苗的目的,前者增殖速度快,后者则再生植株生长势强、驯化移栽易于成活。 The effects of several plant growth regulators(PGRs)were investigated On inducting the protocorm-like bodies(PLBs)and adventitious buds of C. hybridium varieties 'Golden Age', 'Oasis' and 'Waltz'. The results showed that certain concentrations of PGRs could promote the production of PLBs, and the optimal concentrations of PGRs added to MS medium to induce PLBs were4 mg/L BA+ 0. 5 mg/L NAA+ 0. 5 mg/L KT + 0. 2 mg/L TDZ, in which the propagation coefficients of 3 varieties' PLBs were 11.12,9.65 and 7.54 .respectively and were very significantly different from those in the control treatment(4 mg/L BA+ 0.5 mg/L NAA). To establish in vitro regeneration pathways of adventitious buds the 'Waltz' variety whose PLB inductivity was the lowest was selected and the optimal concentrations of PGRs to induce its adventitious buds were 2 mg/L BA + 0.2 mg/L NAA + 1 mg/L PIC(picloram) ,in which the average multiplication coefficient was 5.08. And when 0. 2 mg/L NAA+ 2 mg/L HMI(Hymexazol)was added to MS medium the 4 d rooting percentage could reach 85%. From the above results it was concluded that large scale propagation of C. hybridium could be realized through induction of C. hybridium PLBs and adventitious buds, the former had a high propagation coefficient and the latter had a high growth vigor of regenerated plants which were easy to survive after transplantation.
