摘要
建立了免疫亲和柱净化-柱前衍生化-高效液相色谱荧光检测器测定饲饲料中T-2毒素含量的方法。样品经甲醇-水(体积比80:20)提取,通过免疫亲和柱(IAC)净化,以1-蒽腈(1-anthroylnitrilc,1-AN)为衍生化试剂、4-二甲基氨基吡啶(DMAP)为催化利进行衍生,C1 8色谱柱分离,荧光检测器检测,外标法定量。结果表明:T-2毒素的线性范围为10~200μg/L,加标回收率为76.9%~92.1%,相对标准偏差(RSD)小于6%。该方法可用于饲料中T-2毒素的测定。
A method of determining T-2 toxin in feed was established by high performance liquid chromatography with fluorescence detection via immunoaffinity column purification and precolumn derivatization. A sample was extracted with methanol-water(V/V = 80/20), purified by immunoaffinity columns containing antibodies specific for T-2 toxin, and quantified by reversed phase high perform- ance liquid chromatography with fluorescence detection after derivatization with 1-anthroylnitrile(1- AN)and 4-dimethylam inopyridine(DMAP). The results showed that the linearity range of T-2 toxin was 10-200 μg/L, the recovery was 76.9% -92.1% ,and the relative standard deviation was under 6% ,indicating that this method could precisely determine T-2 toxin in feed.
