探讨HBV -DNA定量和定性聚合酶链反应的意义及其临床实用性。检测 2 83例临床血清标本 ,其中1 2 5例用实时荧光定量PCR测HBV -DNA含量并比较不同乙型肝炎 (乙肝 )病型 ;另 1 5 8例用定性PCR测HBV -DNA并用ELISA法规HBV血清标志物 (HBVM) ,同时进行DNA定量及定性的对比研究。结果示HBV -DNA含量与乙肝病型无明显关系 ;DNA阳性率与乙肝病型及HBVM有关 ;HBV -DNA定量与定性相关性良好。FQ -PCR可真实反映HBV -DNA复制水平 ,有广泛的应用前景 ;定量与定性HBV -DNA检测相关性良好 ,在仅需了解HBV -DNA的复制与否的诊断中 ,可用定性PCR ;对HBVM的意义应重新认识。

To study the quantitative and qualitative PCR detection and clinical application of HBV-DNA HBV-DNA of 283 clinical serum samples were examined Among them,125 samples was tested with fluorescence quantitative PCR and analyzed for the different hepatitis B types,the other 158 samples was tested with routine PCR and HBV markers were tested with ELISA The relationship on the quantitative and qualitative detection of HBV-DNA was studied The quantity of HBV-DNA was not obviously related with hepatitis B types Among the different hepatitis B types and HBV markers,DNA positive rates were different there was identical relation between the quantitative and qualitative PCR of HBV-DNA FQ-PCR can detect the real and accurate quantity of HBV-DNA,but the instrument is so expensive that the application is limited The qualitative PCR can be selected so long as the operating rules can be observed The significance of HBV markers should be rerealized
