摘要
目的:建立壁虎药材中尿苷、腺嘌呤和腺苷的含量测定方法。方法:色谱柱:TSK ODS-100V(4.6mm×250mm,5μm);流动相:A(甲醇)-B(水),梯度洗脱:0-10min(95%B),10-30min(95%B-80%B);检测波长:254nm;流速:1.0mL/min;柱温:30℃。结果:尿苷、腺嘌呤、腺苷的含量分别在0.0568-0.4260μg(r=0.9999)、0.0560-0.4200μg(r=0.9998)、0.1080-0.8100μg(r=0.9999)范围内线性关系良好,回归方程分别为Y=4002769.3526X-4341.3507、Y=4963342.3081X+47345.7427、Y=1153529.8439X+578586.9264,平均加样回收率分别为97.86%(RSD=1.47%)、98.54%(RSD=1.09%)、98.69%(RSD=1.24%),精密度、重复性、稳定性均良好。结论:所建立的方法准确可靠,重复性好,可用于壁虎药材的质量控制。
Objective: To study and establish the quantitative analysis method for uridine, adenine and adenosine in gecko by HPLC. Methods: HPLC was performed on TSK ODS-100V(4.6mmx250mm, 51arn), the mobile phase consisted of methanol and water with gradient at a flow rate of 1.0mL/min. The column temperature was set at 30℃, and UV detection wavelength was at 254nm. Results: Quantitative analysis of HPLC showed that the linear ranges of uridine, adenine and adenosine were 0.056-0.4260lag(r=-0.9999), 0.0560-0.4200lag(r=-0.9998), 0.1080-0.8100lag(r=-0.9999), respectively. The regression equation is Y=4002769.3526X-4341.3507, Y=4963342.3081X+47345.7427, Y-1153529.8439X+578586.9264, respectively. The average recovery of uridine, adenine and adenosine were 97.86%(RSD=1.47%), 98.54%(RSD=l.09%), 98.69%(RSD=l.24%), respectively. Conclusion: The method is accurate, feasible and reliable, therefore which is available for the quality control of gecko.
