土鳖虫醇提物对体外肿瘤细胞增殖的抑制作用及其机制研究 被引量:28

摘要 目的:观察土鳖虫醇提物(ESE)体外抗肿瘤活性及其作用机制。方法:采用MTT比色法、荧光染色技术及流式细胞术,研究ESE对体外培养HepG2和SGC-7901细胞增殖的影响及其对HepG2细胞的诱导凋亡作用。结果:ESE对HepG2和SGC-7901细胞的增殖具有明显的抑制作用,其半数抑制浓度(IC50)分别为0.90μg/mL和0.11μg/mL,并可诱导HepG2肿瘤细胞的凋亡,呈一定的量效关系。结论:ESE可抑制HepG2和SGC-7901细胞的增殖并诱导HepG2肿瘤细胞凋亡,具有较强的体外抗肿瘤活性。 Objective: To investigate the anti-proliferative effects of ethanol extract from Eupolyphaga sinesis Walker (ESE) in vitro. Methods: MTT method was performed to detect the inhibition while AO staining and flow cytometric analysis was performed to confirm the induction of apoptosis against human hepatoma HepG2 cells and human gastric cancer SGC-7901 cells. Results: ICs0 values of ESE were 0.90 and 0.11μg/mL, respectively. After 48h of exposure to ESE, the cells presented the typical morphological changes of apoptosis, and the rate of apoptosis was found to increase in a dose-dependent manner, and the rate of apoptosis was found to increase in a dose-dependent manner. Conclusion: ESE can inhibit HepG2 and SGC-7901 cells proliferation, and induce HepG2 tumor cell apoptosis, which has a obvious anti-proliferative activity in vitro.
