梅毒特异性IgG和IgM检测新方法的临床应用 被引量:1

摘要 目的:应用免疫吸附法建立一种基于明胶颗粒凝集法的梅毒特异性IgG、IgM抗体分型检测方法 ,并探讨其临床应用价值。方法:用羊抗人IgG、IgM抗血清作为吸附剂,对165例梅毒特异性抗体阳性样本分别进行IgG和IgM型抗体的检测,并将检测结果与免疫印迹法进行比较。结果:原倍纯化的羊抗人IgG抗血清对血清样本IgG含量在6.12~34.70 g/L范围时免疫吸附有效,原倍羊抗人IgM抗血清对血清样本含量在2.67~14.68 g/L范围的免疫吸附有效。该方法对165例梅毒螺旋体明胶颗粒凝集(TPPA)试验阳性的标本检测结果与免疫印迹法结果的符合率为100%。结论:经免疫吸附的血清样本用TPPA试验检测梅毒特异性IgG、IgM抗体,可对抗体阳性标本中的IgG和IgM抗体进行简便、可靠的分型检测,有助于梅毒患者的明确诊断、治疗及随访。 Objective To establish a new method for detecting Treponema pallidum specific IgG and IgM antibody based on TPPA and immuno-absorbance method. Methods Using goat anti-human IgG and IgM antibodies as absorbent, 165 TPPA positive specimens were subtyped for Treponema pallidum specific IgG and IgM antibody,and the results were analyzed in comparison with the results of Western-blotting. Results The undiluted goat anti-human IgG antiserum could provide an effective immuno-absorbance when the IgG concentration in the sample was between 6.12-34.70 g/L, and the undiluted goat anti-human IgM antiserum could provide an effective immuno-absorbance when the IgM concentration in the sample was between 2.67-14.68 g/L. The established method had excellent agreement with Western-blotting and the coincidence rate was 100%. Conclusions The new method is reliable and convenient, which could subtype Treponema pallidum specific IgG and IgM antibody in TPPA positive samples. It can provide useful information for the management and follow-up of syphilis patients.
