摘要
目的:建立一去卵巢大鼠模型,通过运动和体外补充雌激素,探讨运动和雌激素对去卵巢大鼠脂肪代谢中GSK-3β、P-GSK-3β的影响。方法:以雌性去卵巢SD大鼠为研究对象,进行为期15周的实验,随机分为假手术安静组(Sham)、假手术运动组(Sham+E)、去卵巢安静组(OVX)、去卵巢运动组(OVX+E)和去卵巢雌激素组(OVX+E2)。运动组大鼠18 m/min,坡度5°,45 min/d,每周运动5 d;去卵巢激素组大鼠每周按体重25μg/kg注射E2,每周3次。实验结束后测定大鼠腹腔内脂肪重量,采用放射免疫方法测血清中雌激素E2水平,Western Blot测定大鼠脂肪总蛋白含量和脂肪组织GSK-3β和P-GSK-3β蛋白表达。结果:1)经过15周实验后,去卵巢安静组大鼠体重显著高于假手术安静组(P<0.05),运动和补充雌激素之后体重明显下降(P<0.05);2)去卵巢安静组大鼠腹腔内脂肪重量最大,运动和雌激素补充干预后脂肪重量明显降低(P<0.05);3)大鼠去卵巢之后脂肪组织中甘油三酯含量显著升高(P<0.05),运动和补充雌激素后又显著下降(P<0.05);4)运动训练后,假手术运动组大鼠和去卵巢运动组大鼠脂肪组织P-GSK-3β/GSK-3β蛋白表达量均显著升高(P<0.05)。结论:1)运动训练和体外补充雌激素,在一定程度上可以缓解去卵巢大鼠腹腔内脂肪的积累;2)中等强度运动训练能够提高去卵巢大鼠体内雌激素水平;3)运动能够上调脂肪组织中P-GSK-3β/GSK-3β蛋白的表达;体外补充雌激素对P-GSK-3β/GSK-3β蛋白的表达却无明显影响。
Objective: The present thesis designed an ovariectomized rat model, through the exercise and estrogen replacement therapy, to explore the effects of exercise and estrogen on GSK -3β, P-GSK -3β of lipid metabolism in ovariectomized rats. Methods: The ovariectomized female Sprague Dawley rats, provided exercise and estrogen replacement for a period of 15 weeks, were randomly divided into five groups: Sham, Sham + E, OVX, OVX + E and OVX +E2. The exercise groups would performed 18m/min, gradient of 5 degree, 45 min and five days a week; The OVX + E2 group was injected the estrogen according to the weight 25 ug/kg weekly, three times a week. After the experiment, the researchers measured the intra-abdominal fat mass in rats, and measured serum levels of estrogen (E2) by radio-immunitied. The total protein content and adipose tissue GSK -3β and P-GSK - 3[3 protein were determined through BCA and Western blot. Results: 1 ) After 15 - week experiment, the body weight in OVX was significantly higher than Sham (P 〈 0.05), and after exercise and estrogen replacement thera- py it decreased respectively(P 〈 0.05 ). 2 )After ovariectomized, the intra-abdominal fat OVX rats were the heavi- est in all group, and through exercise and estrogen replacement, fat mass were decreased significantly (P 〈 0.05). 3 ) The triglyceride content in OVX adipose tissue was significantly higher ( P 〈 0.05 ), after exercise and estrogen replacement therapy the triglyceride significantly decreased (P 〈0.05). 4) The exercise group P-GSK/ GSK protein expression in adipose tissue were significantly higher than quiet group ( P 〈 0.05 ). Conclusion : 1 )Exercise and estrogen replacement therapy in vitro can certain release the ovariectomized rat intra-abdominal fat accumulation. 2) Moderate-intensity exercise can improve the ovariectomized rats estrogen (E2) levels. 3 ) Exer- cise increase adipose tissue P-GSK -3 β/GSK -3β protein expression, but estrogen couldn't influence the P-GSK - 3 β/GSK - 3 β protein expression.
