应用组织芯片检测新疆哈萨克族食管癌核黄素转运基因2蛋白表达及意义

摘要 为检测新疆哈萨克族(哈族)食管鳞癌中核黄素转运基因2(hRFT2)蛋白表达,以探讨hRFT2蛋白在新疆哈族食管鳞癌中的意义。应用组织芯片技术结合免疫组织化学Envision法染色,检测新疆哈族石蜡包埋食管鳞癌组织119例及60例癌旁非癌组织中hRFT2蛋白表达。结果显示,(1)60例哈族癌旁非癌组织中,61.7%hRFT2蛋白细胞核阳性表达伴有少量细胞浆阳性的表达,38.3%阴性表达;在119例食管鳞癌组织中,95.0%出现细胞浆或细胞膜的弥散阳性表达,其余5.0%阴性;(2)食管鳞癌高分化、中分化及低分化三组间hRFT2蛋白表达的阳性率有逐渐降低的趋势,但其三组间hRFT2蛋白表达差异没有统计学意义(P>0.05);(3)按发病年龄、性别、临床TNM分期与淋巴结转移等临床病理特征分组的新疆哈族食管鳞癌中,hRFT2蛋白表达差异没有统计学意义(P>0.05)。由此可知,在新疆哈族食管鳞癌中hRFT2蛋白的表达异常,主要表现为蛋白从细胞核到细胞浆/膜的转位,其异常表达可能与食管癌的发生有关。 To investigate the expression of human riboflavin transporter 2 (hRFT2), and its relationship with esophagus squamous cell carcinoma (ESCC). Methods: Using tissue chip and immunohistochemical Envision method, we selected 119 ESCC cases and 60 non-can- cerous adjacent tissues(NCAT ), and all of them were paraffin-embedded. (1)In 60 cases NCAT, the expression of hRFT2 was found 37 (61.7 % ) in nucleus and little cytoplasm positive, other 23 (38.3 % ) showed negative And in 119 ESCC, i 13 ( 95 % ) showed cytoplasm positive,other 6 (5 %)showed negative. (2)Among the three groups(good, moderate and poor differentiation), the expression rates of hRI"T2 were % 96.7 %, 96.0 %, 85.7 %, respectively; ( 3 ) The expression of hRFT2 in ESCC didn't correlate with sex, age, TNM stage and lymph node metastasis(P〈0.05). The expression of hRVI'2 in Xinjiang Kazakh ESCC is abnormal,mainly including the transloca- tion from nucleus to cytoplasm/membrane,which may relate to the carcinogenesis of ESCC.
