摘要 为研究三氧化二砷(As_2O_3)节拍化疗对HepG2荷瘤裸鼠血管生成拟态(vasculogenic mimicry,VM)的影响,并探讨可能的机制。采用构建HepG2肝癌荷瘤裸鼠模型的方法,用As_2O3_对荷瘤动物模型进行节拍化疗,观察治疗效果及其毒副作用;用HE、过碘酸-雪夫反应(糖原染色,PAS)检测血管生成拟态(VM),免疫组织化学法检测瘤体组织CD31的表达。结果显示,肝癌HepG2细胞在活体模型中可表达内皮相关分子CD31,形成PAS染色阳性、类似血管的管腔样结构。As_2O_3节拍化疗组血管生成拟态受到抑制,而最大耐受剂量组却无此作用。由此可知,As_2O_3节拍化疗可抑制荷瘤裸鼠肝癌的发生发展。 To examine the effects of metronomic chemotherapy with As_2O_3 on vasculogenic mimicry(VM) formations in nude mice bearing HepG2 liver cancer cells and to explore the possible mechanism.The model of HepG2 liver cancer cells tumorbearing nude mice was established,As_2O_3-based metronomic chemotherapy tumor-bearing model was performed.The efficacy and adverse reaction were observed.The number of VM was counted by HE and periodic acid Schiff(PAS) staining.The expression of CD31 was analyzed by immunohistochemistry.HepG2 liver cancer cells expressed CD31 endothelium-related molecules and formed PAS-positive vasculogenic mimicry.As_2O_3-based metronomic chemotherapy group markedly inhibited the formation of VM.But As_2O_3 at maximum dosage had no inhibitory effects on VM formations.As_2O_3-based metronomic chemotherapy can inhibits VM formations by HepG2 liver cancer cells.
