摘要
干苔藓植物标本、多年保存苔藓植物标本等的基因组DNA提取是制约苔藓分子生物学研究的难题。本论文采用常规CTAB、高盐CTAB、改良CTAB、试剂盒、SDS法和快速提取法这6种方法来提取保存多年的苔藓标本干样的基因组DNA。结果表明,改良CTAB法是适于提取苔藓干样基因组DNA的最佳方法,提取的基因组DNA总量较多且杂质较少。进一步的ISSR分子标记实验结果表明,基于改良CTAB法所提取的苔藓标本基因组DNA能有效用于ISSR分子遗传学研究。
The isolation methods of high-quality genomic DNA of dry Moss plants such as rock samples and years-preserved specimen is one of the key difficulties restricting its researches of molecular biology. Six main methods including common CTAB, high-salt CTAB, improved CTAB, Genome Kit, SDS and fast extraction were used to isolate genomic DNA of dry Moss specimen for many years. The results show that the improved CTAB is the optimal method to isolateMoss specimen genomic DNA, whose total yield is higher and impurity is lesser. The ISSR analytical results show that the genomic DNA isolated by improved CTAB method is suitable for the ISSR-based molecular genetics.
