微囊化猪脉络丛细胞脑内移植改善帕金森病样猴异常行为的研究

摘要 目的观察微囊化猪脉络丛细胞脑内移植对偏侧帕金森病(PD)样猴的行为的影响。方法以海藻酸钠-多聚鸟氨酸-海藻酸钠(A-Plo-A)为材料包裹猪脉络丛细胞(CP)。右侧颈内动脉注射甲基-苯基-四氢嘧啶(MPTP)诱发PD样猴模型。采用尾状核定位穿刺、微囊化CP和空白微囊植入PD样猴右侧尾状核区域,观察PD样猴移植术后行为变化。结果微囊化CP组移植后PD样猴帕金森病症状明显减轻,阿扑吗啡(Apo)诱发的异常旋转次数减少(P<0.05),异常行为纠正效果在移植后6个月仍存在。空白微囊组及手术对照组异常行为无变化(P>0.05)。结论 PD样猴脑内移植微囊化猪CP后异常行为明显改善,微囊化PC脑内移植有望成为临床治疗PD的有效方法之一。 [Objective ] To examine the effects of xenografts of microencapsulated bovine choroid plexus cells (PC) on the rotational behavior of hemiparkinsonian monkey recipients. [ Methods ] The hemiparkinsonian model in mon- keys was induced by a unilateral intracarotid injection of methyl-phenyl-tetrahydropyridine (MPTP). Neostriatum punture and PCs microencapsulated in alginate-poly-alginate (APA) membranes, as well as empty microencapsules were grafted into the neostriatum of the hemiparkinsonian monkeys. [Results] After the transplantation the hemi- parkinsonian symptoms subsided and the number of rotations induced by apomorphine decreased (P 〈0.05) for up to 6 months in the group of recipients grafted with microencapsulated PCs. No change was observed in the recipients of empty and operation control group (P 〉0.05). [ Conclusion l The results clearly indicate that the transplantation of microencapsulated PCs can improve the parkinsonian symptoms for long periods of time in hemiparkinsonian monkeys, microencapsulated bovine PC brain transplant is expected to be one of the effective the clinical treatment of PD.
