紫杉醇联合热疗治疗小鼠鼻咽癌CNE-2移植瘤的实验研究 被引量:1

摘要 目的观察紫杉醇(Paclitaxel)联合热疗(hypertherm,HT)对鼻咽癌细胞株CNE-2小鼠移植瘤的抗肿瘤效果,为临床应用提供实验依据。方法采用BALB/c小鼠作为移植性肿瘤实验动物模型。同性别,体重17-20 g,接种人鼻咽癌CNE-2细胞后随机分组,实验设紫杉醇对照组、紫杉醇10 mg/kg组、紫杉醇20 mg/kg组、热疗对照组、热疗组及紫杉醇20 mg/kg﹢热疗联合组,共6组。每组9只。采用一次性小鼠腹腔注射给药方式;紫杉醇对照组小鼠腹腔注射等量生理盐水;并恒温43℃对热疗组肿瘤部位进行热导向化疗,联合治疗组以20 mg/kg紫杉醇剂量一次性小鼠腹腔注射给药,给药60 min后,经水浴局部加热使肿瘤温度达到43℃保持30min,单独热疗组也经水浴局部加热使肿瘤温度达到43℃保持30 min,热疗对照组加热37℃保持30 min。动态观察并测定肿瘤的瘤质量和抑瘤率,评价治疗效果。结果紫杉醇与热疗均可影响鼻咽癌肿瘤的生长速度,使肿瘤缩小,瘤体重量减轻,抑瘤效果较明显;紫杉醇20 mg/kg+热疗联合组抑瘤率与单独应用紫杉醇或单独热疗相比,能显著增强对鼻咽癌小鼠移植性瘤的抗肿瘤效果(P〈0.01);光学显微镜和电镜下可见细胞损伤及凋亡细胞。结论紫杉醇对鼻咽癌移植性肿瘤具有明显的增殖抑制及诱导细胞凋亡作用,且与热疗联合应用能显著增强抗肿瘤效果,为鼻咽癌的临床治疗提供了新的思路。 [ Objective ] To observe anti-tumor effect of paclitaxel combined with heat therapy (hyperthermia, HT) for nasopharyngeal carcinoma (NPC) in CNE-2 cell inoculated mouse and to provide the evidence for clinical application. [Methods] The BALB/c mice, same sex, weight 17-20 g, were randomly divided into groups after inoculating tumour cell, 9 mouse in every group, the experiment included 6 groups: Taxol control group were injected of the same amount saline at one-time intraperitoneal injection way, constant temperature and heat therapy for tumor site by thermal oriented chemotherapy; The combination group were given 20 mg/kg of taxol one-time by intraperitoneal injection after 60 minutes, the local water heating temperature 43~C tumor lasts 30 minutes, heat therapy alone the local heating water of tumor to keep temperature 43~C 30 minutes, heat treatment of the control group lasts 30 min- utes at 37℃. Dynamic observation and determination of the quality and inhibition rate of tumor were used to evaluate the efficacy of the treatment. [ Results ] Paclitaxel and heat treatment of all groups can affect the growth of tumors nasopharyngeal carcinoma (NPC) and shrink tumors , pseudoaneurysm weight or tumor effect is obvious; 20 mg/kg of paclitaxel with heat treatment showed more significantly anti-tumor effect compared with paclitaxel alone or heat treatment (P 〈0.01); Optical microscope and electron visible cell injury and the apoptotic cells. [Conclusion] Taxol inoculated tumor has the obvious nasopharyngeal carcinoma (NPC) proliferation inhibition and induces the apoptosis of function. The combined heat therapy significantly increases antitumor effect and provides new ideas for the clinical treatment of nasopharyngeal carcinoma (NPC).
