摘要
目的初步探讨小白菊内酯及顺铂对人结肠癌细胞SW620细胞凋亡影响的机制。方法培养SW620细胞,单独使用5μmol/L顺铂或10μmol/L小白菊内酯处理SW620细胞,并5μmol/L顺铂和10μmol/L小白菊内酯联合作用SW620细胞。处理24 h后收集细胞,提取总RNA,处理48 h后收细胞提取总蛋白;采用荧光定量PCR和Western Blot的方法检测凋亡相关基因Bcl-2、Bax、Caspase 3、Caspase 9和PARPmRNA水平和蛋白水平的表达变化。结果小白菊内酯及顺铂单药作用时可上调Bax、Caspase 3、Caspase 9和PARP mRNA水平和蛋白水平的表达量,下调Bcl-2 mRNA水平和蛋白水平的表达量,联合作用时对凋亡基因的影响作用更加明显。结论小白菊内酯及顺铂对SW620细胞凋亡的影响可能通过调节凋亡相关基因Bcl-2、Bax、Caspase 3、Caspase 9和PARP的表达发挥作用。
[ Objective ] To investigate the possible mechanism of parthenolide and cisplatin on apoptosis of on SW620 cells. [Methods] After treated with 10 p.mol/L parthenolide and (or) 5 μmol/L cisplatin, the SW620 cells were harvested at different time points for different experiments. After treated 24 h, the total RNA was extracted for quantitative real-time PCR(Q-PCR) assays. And the total proteins of SW620 cells were also obtained after treated 48 h. The expression patterns of apoptosis-related genes under the treatments of parthenolide and (or) cisplatin were analyzed by Q-PCR and Western blot. [Results] The results of Q-PCR and Western blot analysis showed that ex- pression level of Bax, Caspase 3, Caspase 9 and PARP were positively regulated by parthenolide and cisplatin. And the expression pattern of Bcl-2 was down-regulated by parthenolide and cisplatin. Compared to parthenolide or cisplatin alone,significant changes of expression level were observed in the combined treatment. [ Conclusion ] The apoptosis induced by parthenolide and cisplatin in SW620 cell may probably be related to the expression changes of Bcl-2, Bax, Caspase 3, Caspase 9 and PARP.
