青年男性肥胖者相关血浆蛋白标志物的筛查 被引量:2

Screening of protein markers on the plasma of obese young men

摘要 目的利用脱胶电泳一差异蛋白质组学法,筛选与青年男性肥胖相关的蛋白标志物。方法选择10例18~44岁男性肥胖者,结合年龄、性别等因素进行1:1配对对照组,采集其空腹8h以上的静脉血2ml;血浆样品经高特异性的免疫亲和法去除白蛋白和IgG后,进行溶液酶切,对多肽混合样品进行脱胶电泳;利用二维纳流液相色谱-芯片-四级杆飞行时间质谱系统对按等电点分离的多肽样品进一步分离和质谱鉴定,比较两组样本中的差异蛋白质。结果肥胖组与对照组分别分离鉴定出了332、301种蛋白质。两组共有43种差异蛋白质,其中对照组有特异蛋白质17种,肥胖组有特异蛋白质26种。蛋白注释结果表明:与对照组血浆蛋白质相比,肥胖者血浆中脂联素水平较低,而C反应蛋白以及3种磷酸酶的水平较高。结论脂联素、C反应蛋白及3种磷酸酶与青年男性肥胖密切相关。 Objective To screen obesity-related protein biomarkers of young men using differential proteomic method and OffGel electrophoresis. Methods Ten male obese volunteers with the age of 18 - 44 years were selected. The control group was matched with the ratio of 1 : 1 considering the factors of age and gender etc. Two milliliter venous blood was collected after 8 hours fasting. Albumin and IgG were removed from the plasma samples with highly specific immune-affinity method. Then the peptide-mixed samples were separated by pI with OffGel electrophoresis after solution digestion. Further separation and identification were performed by Nano HPLC-Chip-MS/MS system. The different proteins between the two groups were compared. Results Overall, 332 and 301 proteins were identified in the obesity and control groups, respectively. There were 43 proteins with significant differences between the two groups, 17 of which were in the control group and 26 in the obesity group. Protein function annotation results showed that tile level of adiponeetin was lower while the level of C-reaction protein and three other phosphatases were higher in the obesity group compared with the control group. Conclusion Adiponeetin, C-reaction protein and three phosphatases were closely related to obesity of young men.
