摘要
目的探讨丹参酮ⅡA磺酸钠(Sodium TanshinionⅡAsilate)对大鼠肢体缺血再灌注后白细胞介素(IL)-8、肿瘤坏死因子(TNF)-α的影响。方法将60只雄性SD大鼠随机分成对照组、缺血再灌注组和丹参酮组,建立大鼠肢体缺血再灌注损伤模型。利用光镜和电镜观察肺组织形态学改变和超微结构改变,利用双抗体夹心ABC-ELISA法测定血浆、肺组织、骨骼肌中白细胞介素(IL)-8、肿瘤坏死因子(TNF)-α,并进行比较。结果与缺血再灌注组相比,丹参酮组肺血管扩张、炎性细胞聚集、组织水肿等均减轻,线粒体水肿、空泡变性,内膜下基底膜水肿、内皮细胞肿胀等情况明显减轻;血浆、肺组织、骨骼肌中IL-8、TNF-α的含量明显降低,差异均有统计学意义(P<0.05)。结论丹参酮ⅡA可以降低大鼠肢体缺血再灌注后血浆、肺组织、骨骼肌中IL-8、TNF-α的水平。
Objective To investigate injury in rats and the change of IL-8 and protective effect of Tanshinion II A (tan TNF-ain serum, lung tissue and skeletal ]/A) on ischemia-reperfusion lung muscle after ischemia-reperfusion. Methods Referring to normal method , the model rats which underwent 4 hours ischemia and 2 hours reperfusion of hind limbs were made. 60 $D rats were randomly divided into 3 groups: the control group, the limb- ischemia-reperfusion group,the Tanshinon group. The histological and ultra-structure changes were observed under light microscope and electron microscope, respectively. We observed changes of IL-8 and TNF-a in tung tissue of different groups. Results 1 The damage in lung tissues of group B was more seriously than that of group A and group C under optical microscopy and e-lectron microscope. 2 After hind limbs suffered iscbemia-reperfusion, the level of IL-8, TNF-a in serum and lung tissue were much higher in group B than group A; IL-8,TNF-a in serum or skeletal muscle and lung tissue were much lower in group C than group B. Conclusion Tanshinone IIA can alleviate lung ischemia-reperfusion injury in rats.
