摘要
氟烷基因(RYR1基因)是使猪产生劣质肉及应激综合征的主要因素之一。本研究采用PCR-RFLP技术对45头可乐猪RYR1基因的多态性进行了检测,并通过直接测序对检测结果进行了验证。结果表明:试验猪群体中仅存在显性纯合(HalNN)和杂合(HalNn)两种基因型,不存在隐性纯合基因型(Halnn)。其中,HalNN为优势基因型,频率为0.9111;HalN为优势等位基因,频率为0.955 6。同时,在可乐猪群体中,RYR1基因的变异位点为低度多态(PIC<0.25),并偏离了Hardy-Weinberg平衡(P<0.05)。本研究结果为可乐猪的选育及开发利用提供了理论依据。
RYR1 is one of the main candidate genes for pock of poor quality and porcine stress syndrome. In this study, RYR1 gene variants of 45 Kele pigs were detected by PCR-RFLP, and test data were validated by direct sequencing. Results showed that two genotypes (Hal^NN and Hal^Nn) except for Ha^nn were found, Hal^NN was advantage genotype and frequency was 0. 9111, HalN was advantage allele and frequency was 0. 9556, while the variant in RYR1 gene was low polymorphie and unfitted Hardy-Weinberg equilibrium withx2-fitness test in Kele pigs. The obtained data provided a basis for breeding and exploitation of Kele pigs.
