醋柳黄酮对家兔心肌缺血再灌注损伤的保护作用

摘要 【目的】观察醋柳黄酮对结扎家兔冠状动脉前降支所致的心肌缺血再灌注家兔心肌组织中Bcl-2及Bax基因及蛋白表达的影响。【方法】选用24只健康新西兰大白兔随机分为假手术组、模型组、醋柳黄酮组(剂量为0.96 mg.kg-1.d-1)。采用结扎家兔冠状动脉前降支方法复制急性心肌缺血模型,Real-time PCR及Western blot方法检测Bcl-2及Bax基因及蛋白表达变化。【结果】模型组心肌组织Bcl-2、Bax mRNA,Bax蛋白表达显著升高(均P<0.01),Bcl-2蛋白表达显著降低(P<0.01);醋柳黄酮组可显著升高Bcl-2 mRNA及蛋白表达,降低Bax mRNA与蛋白表达,与模型组比较差异有统计学意义(P<0.05)。【结论】醋柳黄酮对家兔冠脉结扎后引起的心肌缺血损伤的保护作用机制可能与其对凋亡基因的调节密切相关。 Objective To study the effect of sindacon on bcl-2 and bax protein and gene expression in myocardial tissues of rabbits with myocardial ischemia and reperfusion induced by ligation of anterior descending coronary artery. Methods Twenty-four rabbits were randomly divided into sham operation group, model group and sindaeon group ( in the dosage of 0.96 mg · kg 1 . d- 1 ) . Rabbit model of myocardial ischemia and reperfusion was induced by ligation of anterior descending coronary artery. Western blot and real-time PCR methods were used to detect the expression of bcl-2 and bax. Results The expression levels of Bcl-2 and Bax mRNA as well as Bax protein were increased, and Bcl-2 protein was decreased obviously in the myocardial tissues of model group ( P 〈 0. 01 compared to the sham operation group). Sindaeon had an effect on increasing Bcl-2 mRNA and protein expression, and on decreasing Bax mRNA and protein expression in the myocardial tissues of model rabbits ( P 〈 0.05 compared to the model group ). Conclusion Sindacon has protective effect on rabbits with myocardial ischemia and reperfusion injury, and the protective mechanism may be related to the regulation of apoptosisassociated genes.
