摘要
【目的】筛选青天葵种子无菌萌发的适宜条件。【方法】选取不同生长时期的青天葵果实,观察接种时间,光照条件,基本培养基(1/2 MS、MS、MB2),激素6-苄基嘌呤(6-BA)、α-萘乙酸(NAA)以及天然添加物对青天葵种子萌发与生长的影响。【结果】青天葵种子在黑暗条件下能萌发,在MS+3 mg/L 6-BA+体积分数10%苹果汁或1.0 g/L酵母膏培养基上,青天葵种子萌发率高且长势良好。【结论】建立了青天葵种子无菌萌发条件,为青天葵人工繁育提供新途径。
Objective To optimize the growth conditions for the aseptic germination of Nervilia fordii seeds. Methods Nervilia fordii seeds collected at different growth periods were used for the experiment. The influences of inoculation time, illumination time, culture medium (1/2 MS, MS and MB: ), hormones of 6-benzyladenine (6- BA) and a-naphthaleneacetic acid ( NAA), and natural additives on the seed germination and seedling growth were observed. Results The germination of Nervilia fordii seeds occurred under dark and aseptic cultured conditions, and the seeds had a high germination rate and grew well on 1.0 g/L yeast extract medium or on MS medium with 3 mg/L of 6-BA and 10% of apple juice added. Conclusion The aseptic germination conditions for Nervilia fordii seeds have been optimized successfully, which will supply a new way for the breeding of Nervilia fordii.
