T-7-RGD肽对人非小细胞肺癌(A549)和人脐静脉内皮细胞(ECV304)的影响 被引量:1

摘要 目的:研究改造后肿瘤抑素7肽(T-7-RGD肽)对人非小细胞肺癌(A549)和人脐静脉内皮细胞(ECV304)活性的影响。方法:采用生物合成技术合成了T-7-RGD肽,高效液相和质谱分析显示纯度为98.67%;通过MTT法、生长曲线法、HE染色等实验初步观察T-7-RGD肽对人非小细胞肺癌(A549)和人脐静脉内皮细胞(ECV304)增殖抑制作用和形态影响。结果:由MTT实验和生长曲线实验得出,人非小细胞肺癌细胞A549和人脐静脉内皮细胞ECV304半数抑制率IC50分别为150.34μg/ml和420.81μg/ml,且呈剂量、时间依赖性。T-7-RGD肽对A549作用明显,改变了其生长曲线的基本走势,而对人脐静脉内皮细胞只是产生了数量上的改变,生长曲线基本走势没有改变。HE染色后人非小细胞肺癌A549实验组与对照组相比形态学发生了明显的变化,而人脐静脉内皮细胞ECV304变化不明显。结论:T-7-RGD肽可抑制人非小细胞肺癌细胞A549生长,促进其凋亡,而对内皮细胞的作用不明显。 Objectlve:To study the effect of biological activity of T - 7 - RGD peptide(RGDSGGCNYYSNS) on the cell proliferation and apeptosis of non - small cell lung cancer( A -549) and human umbilic:d vein endothelial cell( ECV - 304) Methods:T -7 - RGD peptide was synthesized and its purity is 98.67% by high - performance liquid chromatography. The effect of T-7 -RGD peptide on inhibiting proliferation and morphology of non- small cell lung cancer( A- 549)and human umbilical vein endothelial cell( ECV -304)was observed by MTT, cell growth curves and HE staining. Results:By MTT and growth curve experiments, IC50 of A - 549 and ECV - 304 cells were 150. 34μg/ml and 420. 81μg/ml respectively, showing dose - and time - dependent. T - 7 - RGD peptide inhibited the groWth of A - 549 obviously and Change the basic trend of cell growth curve. However,the growth trend of human umbilical vein endothelial cells(ECV304) was consistent with the blank control group, only showing the decrease of the cell number. A -549 cells was obviously changed by HE staining, which consistent with the blank control group. The effect of T - 7 - RGD peptide o, human umbilical vein endothelial cells ( ECV304 ) was not apparent. Conclusion :T- 7 -RGD peptide can inhibit the proliferation of A -549 cells and promote apoptosis. The effect on ECV304 cells was not apparent.
