摘要 目的研究探讨微波辐射的非热效应对人肺癌A549细胞损伤的反应,以期为微波治疗肿瘤提供更全面的理论依据。方法采用剂量为100W(12mv/cm2)、150W(18mv/cm2)和200W(21mv/cm2)的微波辐射冰浴方法制备的非热效应细胞模型10min;24h后收集细胞,以Western印迹方法分析细胞Cytochromec、Caspase-3,GRP78、Caspase-4,CathepsinD的表达;MDC染色、激光共聚焦显微镜观察细胞溶酶体变化。结果辐射后A549细胞Cytochromec、GRP78蛋白表达增加,并且凋亡相关蛋白Caspase-3,Caspase-4活化;细胞溶酶体活性增强表现为CathepsinD表达增加以及MDC染色荧光强度加强。结论微波辐射非热效应对组织细胞所造成的损伤反应可能是通过细胞线粒体、内质网和溶酶体途径共同所诱导的细胞凋亡,为微波治疗肿瘤提供新的思考方向。 Objective To investigate the injury on Cell Line A549 induced by non-thermal effects of Microwave Radiation, in order to provide theoretical basis for Microwave treatment of cancer. Methods Ice bath was employed to establish microwave radiation non-thermal effect model, and dosages were 100 W(12mv/cm2), 150 W-18mv/cm2 and 200 W(21mv/cm2) for 10 min. After 24 h, Western Blotting was used to detect the expressions of cytochrome c, caspase-3, GRP78, caspase--4, cathepsin D proteins. Laser Scanning Confocal Microscope was used to dectect the changes of lysosomes. Results The non-thermal effect of microwave radiation on human A549 cells could induce the release of mitochondfial cytochrome c, and induced activation of caspase-3 and easpase-4, and upregulated the expression of GRF78 and eathepsin D, and enhanced acidification of lysosomes. Conclusion The injury on Cell Line A549 induced by non-thermal effects of Microwave Radiation may be apoptosis through mitochondrial , endo- plasmic reticulum and lysosome pathways together, offering new thinking direction for microwave treatment of cancer.
